The effect of NTP on β-catenin expression in confluent HaCaT cells. (a) The results of Western blot and RT-PCR. Confluent HaCaT cells were treated with NTP for 5 minutes at 3, 6, 9, 24 hours before harvesting. (b) Confluent HaCaT cells were treated with NTP for the indicated length of time and incubated for 6 hours. After harvesting the cells, the total protein extracts were used for Western blot analysis. GO: Gas only (argon). (c) One day after seeding, HaCaT cells were transfected with scrambled siRNA (sc) or siRNA against β-catenin. Six hours before harvesting, cells were treated, or not, with NTP for 5 minutes. After harvesting, the total RNA and protein extracts were used for RT-PCR and Western blot analysis, respectively. Data shown are representative of three independent experiments. GAPDH was used as loading control.
