Herc5 expression is significantly reduced by CYP1B1 in HeLa cells. (A) RT-PCR analyses. Cells were transfected with pcDNA3.1( zeo)-CYP1B1 (5 μg) or CYP1B1 specific siRNA (40 nM) for 48 hr. Total RNA was isolated and the Herc5 gene was amplified with specific primers. Expression of GAPDH mRNA was measured as an RNA control. (B) Western blot analyses. Cells were transfected with pcDNA3.1(zeo)-CYP1B1 (5 μg) or CYP1B1 specific siRNA (40 nM) for 48 hr. Cell lysates were isolated and western blot analyses were performed against Herc5. β-Actin was used as an internal loading control.
