Chloroquine further sensitises Tsc2−/− MEFs to nelfinavir‐induced ER stress. Tsc2−/− and Tsc2+/+ MEFs were treated with DMSO vehicle alone, 20 μM nelfinavir (NFV), and/or chloroquine (CQ, 10 μM and 20 μM) as indicated. A) Following 3 h drug treatment, protein lysates were subjected to western blot analysis to determine levels of total and phosphorylated ‘(P)‐rpS6’, total and phosphorylated ‘(P)‐S6K1’, LC3‐II, TSC2 and β‐actin. B) CHOP mRNA was analysed and standardised against β‐actin (n = 3, *p < 0.05).
