LPS-treated DCs modulate development and differentiation of Treg subpopulations mediated by CD127 and 3G11. Splenocytes were isolated from mice which were i.v. transferred into LPS-treated DCs-MOG+LPS or DCs-MOG and DCs without incubation with LPS and MOG peptide shown in Fig. 3. Cells were re-stimulated with mouse IL-2 (1 ng/ml) and MOG peptide (0.1 µM) for 72 h at 37 °C. T lymphocytes were stained by anti-mouse CD4, CD25, CD127, GITR, FoxP3, and 3G11 antibodies. CD4+CD25+FoxP3+GITR+ Tregs were gated. Protein expression of 3G11 and CD127 on CD4+CD25+FoxP3+GITR+ Tregs was detected using flow cytometry.
