Expression of the desired DENV2 proteins in rhesus fibroblast cultures after infection with RRV-DENV2 recombinants. RF cell lysates were prepared, and cell culture supernatants were harvested. Proteins were separated by SDS-polyacrylamide gel electrophoresis, and the separated proteins were transferred onto a PVDF membrane and subsequently tested with DENV2-specific antibodies. (A) Recombinant RRV-E85 (rRRV-E85) represents either the cell lysate or the cell culture supernatant from RF cells infected with RRV-E85. Blots were probed with an antibody specific for the E protein of DENV2 (4G2). (B) Recombinant RRV-NS5 represents cell lysate from RF cells infected with RRV-NS5.
