Expression of surface CD4 and CD8 (top row) in 1-week-old neonate (a) and 6-week-old adult (b) thymi of IP3R-TKO and control mice. Total thymic cellularity is shown above plots (mean±s.e.m.). CD8+CD4 thymocytes were subgated (red boxes) and examined for surface expression of CD24 and CD3 to observe ISP cells contained in the CD8+ CD4 populations shown in a and b. This was done in both neonatal (c) and adult (d) thymi of IP3R-TKO and control mice. Numbers in plots indicate percentage of cells. Absolute cellularity as depicted in ad for neonatal (e) and (f) adult thymic subsets (mean±s.e.m.). All animals displaying signs of malignant disease were excluded from these analyses. Data are representative of a minimum of five (af) independent experiments. *P<0.05, measured with two-tailed Student’s t-test.
