RNA was isolated from individual tumour cells obtained from three different mice to generate cDNA. Primers to exon 34 of the mouse Notch1 gene were used to amplify the DNA sequence, which was then subcloned, and 10 clones from each mouse were sequenced. All identified variants are predicted to result in truncations of encoded Notch1 proteins (also refer to Supplementary Fig. 10).
