SALSA consists of two sequential reactions, isopropylamidation and methylamidation. In the first alkylamidation, α2,6- and α2,3-linked sialic acids are selectively converted into their iPA-derivatized and lactone forms, respectively. After the removal of excess reagents by HILIC SPE, a 1% methylamine solution is added to promote alkali-driven lactone cleavage, and solvents were then removed in vacuo.
