Construction of Drosophila Rh7-Cap. (A) Alignment of C-terminal region of Drosophila Rh7 and A. cerana UV light-sensitive visual opsin. The location of Helix VIII in the crystal structure of squid rhodopsin46 is indicated by a box. The amino acid sequence from position 400 to the C-terminal end of Rh7 was substituted with the corresponding amino acid sequence (from position 344 to the C-terminal end) of A. cerana UV light-sensitive visual opsin. (B) Western blot analysis against the C-terminal rho1D4 epitope tag. A clear signal was detected in Rh7-Cap-transfected cells, but not in mock- or Rh7 (full-length)- transfected cells. (C) Photoreaction of Rh7-Cap regenerated with 11-cis-retinal. The black curve was calculated by subtracting the spectrum before irradiation from that after UV light irradiation. The red curve was calculated by subtracting the spectrum after UV light irradiation from that after subsequent yellow light (>500 nm) irradiation.
