Screening of isoform-targeted compounds to identify HIV-1 latency reversing agents (LRAs). Identification of largazoles as potent LRAs. JLAT10.6 cells were incubated with 10 ng/mL of TNFα, 1 μM of SAHA (vorinostat) and 10 μM (a), 1 μM (b), or 0.1 μM (c) of screening compounds for 24 h. The percent of GFP positive cells were analyzed by FACS and the degree of HIV-1 reactivation was normalized to TNF-α. SDL148, JMF1080 and SDL256 are largazole and two analogs respectively. The mean values (mean ± s.e.m.) of a representative experiment in triplicate for each concentration is shown. ***p < 0.001 (ANOVA).
