LDH-A overexpression blocked apoptosis induced by DDP and/or HIF-1α siRNA. (a,b) mRNA and protein expression of HK2, LDH-A, PKM2 and PDK1 was examined with qRT-PCR (a) and western blot (b) in PCa xenografts. (c,d) PC-3 cells were transfected with a human LDH-A cDNA construct or empty vector (pcDNA) for 48 h. Following 24 h of cDNA treatment, the cells were treated by transient transfection with si-HIF-1α alone, DDP alone, or both in combination. Cell lysates were subjected to western blot analysis with the indicated antibodies (c) and an apoptosis ELISA (d).
