FOXQ1 overexpression promotes cell proliferation and FOXQ1 silence causes premature senescence. (a) The 2BS cells expressing the indicated genes and shRNAs were cultured and growth curves were determined by the MTT assay. (b) The 2BS cells were infected with indicated retroviruses and cultured in the 6-well plates for 6 days, followed by fixation and staining with crystal violet. (c and d) Representative images of the indicated cells with stained for senescence-associated heterochromatin foci by DAPI (c) and SA-β-gal activity (d) were shown (Scale bar, 10 μm). (e) The percentage of cells positive for SA-β-gal in each sample was determined. At least 300 cells were counted for each sample. (f) Cell cycle was measured. Values are means±S.D. of triplicate points from a representative experiment, which was repeated three times with similar results. *P<0.05
