Phenotypic analysis in a contemporary clinical isolate. WHO X (WT) and isogenic knockouts, as indicated, were suspended to an optical density at 600 nm (OD600) of 0.1, cultured for 3 h at 37°C, and diluted to 5 × 105 CFU/ml. Cultures were serially diluted and spotted onto a gonococcal base solid medium (GCB) plate, in addition to GCB plates supplemented with 9-aminoacridine (20 μM), bile salts (0.2%), azathioprine (118 μg/ml), or polymyxin B (1,600 U/ml). Plates were incubated at 37°C in a 5% CO2 atmosphere and inspected after 22 h. Experiments were performed in triplicate, and representative images are presented.
