Knockdown of D2HGDH-GAL3ST2 decreased cell proliferation and cell motility in prostate cancer cells. A. Two siRNAs specifically knocked down the fusion RNA D2HGDH-GAL3ST2, with no significant effect on the parental D2HGDH. B. Cell number was dramatically reduced by knockdown of D2HGDH-GAL3ST2 in LNCaP cells. C. Cell proliferation was measured by cell counting (left) and MTT assays (right). D. Cell motility was measured by wound healing assay. E. D2HGDH-GAL3ST2 was enriched in the cell nucleus. The ratio of nuclear vs. cytoplasmic RNA levels of GAPDH, MALAT1, and the fusion were measured by qRT-PCR using RNA extracted from a cell fractionation assay. *, p < 0.05; **, p < 0.01; ***, p < 0.001; ****, p < 0.0001.
