SOD-2 contributed to TNF-α-mediated EMT and migration in A549 cells. (a) The expression of SOD-2 at mRNA and protein level was significant inhibited after transfection with SOD-2 siRNA in A549 cells. (b) After 72 h of TNF-α treatment, the expression of SOD-2, E-cadherin, vimentin, MMP2 and MMP9 in A549 cell transfected with SOD-2 siRNA was detected by Western blot. (c) Representative immunofluorescent micrographs show the expression of E-cadherin and vimentin in SOD-2-knockdown cells. After TNF-α treatment for 48 hours, the migration ability of A549 cell transfected with SOD-2 siRNA was measured by wound healing migration assay (d) and transwell migration assay (e). (f) Cell number was determined by counting the positive cells number in higher field and data was shown as mean ± SD (three experiments, *p < 0.05, compared to control; # p < 0.05, control cells treated with TNF-α compared to SOD-2-knockdown cells treated with TNF-α).
