Cultures of cells expressing cytoplasmic TEV protease (Cyt) or ribosome attached TEV protease (Rib) were grown with or without 100 ng/ml aTc (to induce TEV protease). Samples were taken after 5 h for Western blot analysis with streptavidin-AP. BCCP, biotin carboxyl carrier protein. Detection of biotinylated of FtsQ-PSBT (C) was done as above except that ftsQ was induced with or without 0.2% arabinose.
