CoQ₀ increases Beclin-1/Bcl-2 and Bax/Bcl-2 ratios, and inhibits HER-2/neu/AKT/mTOR signalling in SKOV-3 cells. (A) Dose-dependent effect of CoQ₀ (0–30 µM, 24 h) on changes in Beclin-1, Bax and Bcl-2 proteins were determined by Western blot. Relative changes in the ratio of (B) Beclin-1/Bcl-2 and (C) Bax/Bcl-2 in accordance to dose were quantified by commercially available software, and presented as a histogram, control representing as 1.0 fold. Results expressed as mean ± SD of three independent assays (n = 3), and significant at ***p < 0.001 compared with untreated control cells. (D) Time-dependent effect of CoQ₀ (0–24 h, 30 µM) on phosphorylation of HER-2/neu (Y1221), AKT (Ser473) and mTOR (S2448), and cleavage of PARP were determined by Western blot. Results expressed as mean ± SD of two independent assays (n = 2).
