Effect of Niclosamide on RANKL-induced osteoclast specific gene expression in BMM cultures. Murine bone marrow-derived macrophages (BMM) were pretreated with or without niclosamide for 1 h in medium containing MCSF (30ng/ml) and with RANKL (100 ng/ml) for another 48 hours. Total RNA was isolated with TRIzol, and 1 μg of total RNA was used to synthesize cDNA. cDNA was used as a template for real-time PCR with mouse-specific primers. The relative expression of each target gene compared to β-actin was calculated using the 2-ΔΔCt. Data represent means ± SD of more 3 cultures. *p < 0.05, **p < 0.01, significantly different from values after treatment with DMSO and RANKL.
