Figure 2.
MSI1 increased the aspect ratio and decreased viscoelasticity of GBM cells. (A)(B) MSI1-depleted 05MG cells and MSI1-overexpressed U251 cells were immune-stained for F-actin (Red) and DAPI (Blue) and observed by fluorescent microscopy. The ratio of mean aspect (defined as the ratio of the length of major and minor axes, AR) were shown in the right (n = 100). (C)(D) Fluorescent color maps of F-actin networks distribution in MSI1-depleted 05MG cells and MSI1-overexpressed U251 cells. The formation and the angular distribution of F-actin orientation in MSI1-depleted 05MG and MSI1-overexpressed U251 cells were analyzed by assessing and quantifying the immunofluorescence intensities using MatLab software. The mean of Kurtosis from 45 cells was shown in the right. (E) (F) The intracellular elastic modulus (G’) and viscous modulus (G”, at 10 Hz) of MSI1-depleted 05MG and MSI1-overexpressed U251 cells were shown as mean ± SEM (n = 100 beads/ 35cells). Each bar chart is presented as mean ± SEM. The experiments were repeated at least three times. The statistical significance was assessed by one-way ANOVA, *P < 0.05 (relative to the control group).
