Expression of the genetic adjuvant and parasite-specific antigens in DCs transduced with the adenoviral vector. Low-titer adenovirus was made with dnSHP as the genetic adjuvant. (A to D) HEK293T cells and dendritic cells were transduced with adenovirus expressing dnSHP (genetic adjuvant) and Tc24 and TSA-1 (antigens). Western blots of cell lysates were probed with anti-HA, anti-SHP, anti-Tc24, and anti-TSA1 antibodies to confirm the expression of the adjuvant, antigen, and fusion protein products. (E) Dendritic cells were transduced with different titrations of the viral particles, and cell lysates were probed with anti-SHP-1 antibody. Doses as low as 100 vp (102 vp) per cell gave good expression of the adjuvant, as confirmed by Western blotting.
