Effects of PKC agonist and antagonists on
nocodazole-induced cell-cell adhesion in NHEK cells. (A) NHEK cells
were treated with DMSO (a), GF109203X at the concentration of 1 μM
(b), and 10 nM PMA (c) for 1 h. PMA itself induced cell-cell
adhesion. (B) Western blot analyses for involucrin and loricrin
expression after PMA treatment. C, NHEK cells were treated with
nocodazole for 48 h (a), PMA for 48 h and then treated with
nocodazole for 1 h (b), PMA and GF109203X for 48 h followed
by nocodazole treatment for 1 h (c), and PMA for 48 h
followed by treatment of GF109203X and nocodazole for 1 h (d). All
cells were visualized by IF staining with anti-E-cadherin. GF10903X
could not reverse the long-term inhibitory effect of PMA on
nocodazole-induced cell-cell adhesion. Bar, 20 μm.
