Perfusion significantly influenced the morphology of neuroblastoma spheroids originated from SY5Y in long-term culture. (a) Representative real-time imaging of neuroblastoma cells SY5Y cultured in Matrigel sandwich comparing the static and perfusion culture over 14 days of culture. Scale bar: 500 µm. (b) Image analysis using ImageJ reveals the significant growth difference at day 14, between static and perfusion culture. Mean spheroid areas were compared as representative parameters. (c) Tissue metabolic viability shows that at day 14 perfusion culture has significantly higher viability compared with static culture. For statistics, all the groups have four replicates and the experiments were repeated for three times. Results are presented as mean ± SD. *p < 0.05 by Student’s t-test.
