Cryptococcus gattii presence alters zinc transporter expression in A. castellanii. Amoebae (1 × 105 cells) were incubated in PYG medium with C. gattii WT or zip1Δ (1 × 106 cells) in 12-well plates for 3 and 24 h, at 30°C. The cells were washed with PBS and RNA from amoebae was isolated and extraction was performed, followed by cDNA synthesis. The measured quantity of zinc transporter mRNA from ZIP coding genes (A) or ZnT coding genes (B) in each sample was normalized using the threshold cycle values obtained for the Actin gene. Data are shown as the mean ± standard deviation from three experimental replicates of three biological replicates.
