Phosphorylation-dependent mobility shift of Mpphot. After desalting of the lysates of blue light (25 µmol photons m−2⋅s−1)-irradiated Mpphot/MpphotKO using an Amicon Ultra 30K device (EMD Millipore), 15 units of FastAP thermosensitive alkaline phosphatase (Thermo Fisher Scientific) was treated for 1 h at 37 °C. The band intensity histograms are shown. The light- and cold-dependent phosphorylations of Mpphot (Fig. 2 F and G) completely disappeared by the phosphatase treatment.
