Inhibition profile of cladosporin, 3-epi-isocladosporin, and isocladosporin for the promastigote growth of WT and genetically manipulated parasites. WT, overexpressor (WT[pLysRS-1+]), heterozygous mutant (LysRS-1/HYG), and add-back (LysRS-1/HYG[pLysRS-1+]) parasites were treated with cladosporin (A), 3-epi-isocladosporin (B), and isocladosporin (C) with concentrations above their IC50s: 5 μM, 65 μM, and 160 μM, respectively. The cell growth was determined after 72 h of drug treatment. In the absence of drug treatment (Untreated), the growth of each parasitic line was normalized to 1.0. After treatment with each drug (Treated), growth was calculated relative to the corresponding untreated control.
