Comparison of an optical flow-derived beat pattern with Ca⁺⁺ oscillations. Brightfield (a) and corresponding Ca⁺⁺ (b) image of cardiomyocytes, recorded at a peak of the Ca⁺⁺ signal. To improve contrast, Ca⁺⁺ sensitive dye fluorescence image was inverted, thus darker areas indicate higher Ca⁺⁺ concentration. Beat patterns (c) were obtained from optical flow analysis, and show a pattern similar to that of the Ca⁺⁺ -dependent total fluorescence (d). Recording framerate: 1/s, field of view: 140 μm.
