Comparison of an optical flow-derived beat pattern with Ca++ oscillations. Brightfield (a) and corresponding Ca++ (b) image of cardiomyocytes, recorded at a peak of the Ca++ signal. To improve contrast, Ca++ sensitive dye fluorescence image was inverted, thus darker areas indicate higher Ca++ concentration. Beat patterns (c) were obtained from optical flow analysis, and show a pattern similar to that of the Ca++ -dependent total fluorescence (d). Recording framerate: 1/s, field of view: 140 μm.
