Binding of PEX5 to PTS1 cargoes does not require ubiquitylation by TRIM37. (A) HepG2 cells were transfected with GFP and myc-PEX5 (WT) (negative control) or GFP-ACOX1 together with myc-PEX5 (WT), (K464A), or (ΔCT51). Cell lysates were collected for GFP-IP and Western blot detection of their association with PEX5. (B) HepG2 control and TRIM37 KD cells were transfected with GFP, GFP-ACOX1, or GFP-PECR. GFP-IP was performed to detect their interactions with endogenous PEX5. (C) HepG2 cells transfected with the indicated constructs were treated with 50 µg/ml CHX in the absence or presence of 10 µM MG132 for 6 h.
