Two perpendicular views of the CREB–CRE complex to illustrate the location of residues that influence CRE/AP-1 specificity. The two basic residues that lower CRE/AP-1 specificity are colored blue, while the four glutamic acid residues that raise CRE/AP-1 specificity are colored red. The conserved quintet is shaded yellow. The residues involved in CRE/AP-1 selectivity all reside on the outside surface of the helix. This observation emphasizes the potential of protein–protein interactions for modulating specificity.
