NK cells enriched from human PBMCs were primed with 200 U/ml IL-2 for 24 h. Cells were treated with medium, 100 nM rapamycin, or 100 nM Torin-1 for 1 h prior to stimulation with plate-bound anti-NKG2D Abs for 5 h. IFNγ production was measured in CD56^bright and CD56^dim NK cells by flow cytometry. (A) Results are shown for one donor and are representative of three donors.
