Effects of the length of splints on the efficiencies of cyclization of l-DNA₆₆ and its polymerization under conventional conditions. All the DNA substrate was added to the mixture all at once at the beginning of the reaction. Each of the splints is complementary to equal number of nucleotides in the 5′- and 3′-ends of l-DNA₆₆, respectively (the binding mode of splint-12 nt is presented in Figure 1A). The reaction conditions: [l-DNA₆₆]₀ = 5 μM; [splint]₀ = 10 μM; 20 U T4 DNA ligase in 1 × T4 ligase buffer at 20°C and 12 h. The sequences of splints were listed in Supplementary Table S1.
