Effects of miR-491 on OS Cell Metastasis and Sensitivity to CDDP Are Mediated through CRYAB
(A) Restoration of CRYAB blocked inhibition of OS cell invasion by miR-491. Saos-2 cells stably expressing miR-491 were transfected with a CRYAB expression plasmid for 24 hr and then subjected to a Transwell assay. (B) Restoration of CRYAB protected cells from combination treatment with miR-491 and CDDP. Saos-2 cells stably expressing miR-491 were transfected with a CRYAB expression plasmid. After 24 hr of transfection, cells were incubated with or without CDDP for 48 hr and then subjected to an MTT assay. (C) Silencing of CRYAB blocked stimulation of cell invasion induced by miR-491 inhibition. Saos-2 cells stably expressing miR-491-antisense were transfected with an siRNA of CRYAB (siCRYAB) for 24 hr and then subjected to a Transwell assay.
