β-Catenin is required for p65 nuclear translocation. (A) Nuclear extracts were used to immunoblot nuclear factor-κB (NF-κB) p65 and β-catenin and normalized against Histone H3. Cells were pretreated with XAV-939 (10 µM) or DMSO for 16 h, after which they were stimulated with IL-1β (1 ng/mL) for 30 min. Data represent six independent experiments. (B) Nuclear extracts were used to immunoblot NF-κB p65 and normalized against Histone H3. Cells were transiently transfected with a green fluorescent protein (GFP) or S33Y-construct for 24 h, and then treated with IL-1β for 30 min. Data represent nine independent experiments.
