Immunotherapy-induced Tregs displayed in vivo suppressive activities that persist after the discontinuation of EPIT but not SLIT or OIT. Mice were sensitized and administered CD4+CD25+ T cells isolated just after or 8 weeks after the end of EPIT, OIT and SLIT. These mice were then compared with non-transferred sensitized mice and naive mice. Splenocytes were stimulated with peanut for 72 h. The concentrations of IL-5 (a), IL-13 (b), IL-10 (c) and IFN-γ (d) were measured in the supernatants using Bioplex. (e) Mice were exposed to oral peanut for 10 days, and the esophagi were harvested. Formalin-fixed, paraffin-embedded tissue sections were stained with hematoxylin–eosin–safranin, and eosinophil infiltration of the esophagus was measured by a double-blind reading in a × 40-powered field. The results are expressed as the median of the number of eosinophils per mm2 with range for each group of mice.
