(A) The mRNA expression of HPA1 was determined using qRT-PCR in SW1990 (left panel) and PANC-1 (right panel) cells with or without 10 μM gemcitabine treatment for the indicated times. Results are representative of three independent experiments. (B) Western blot analysis was used to determine protein abundance of HPA1 after treatment with 10 μM gemcitabine at the indicated time points. (C) BALB/c nude mice bearing SW1990 xenografts were administered a single dose of vehicle (PBS) or gemcitabine (GEM, 50 mg/kg) for either 24 or 48 h. Tumor tissues were obtained and HPA1 mRNA levels were analyzed by qRT-PCR. (D) Immunohistochemistry was applied to detect the expression of HPA1 in xenografts following treatment with gemcitabine.
