Enforced expression of miR-340 inhibits growth and invasion, and induces cell apoptosis in breast cancer cells in vitro. (A) Effect of miR-340 on cell proliferation was measured by MTT assay after NC, mimics or inhibitor transfection in MDA-MB-231 and MDA-MB-468 cells. (B) Colony formation, (C) cell cycle distribution, and (D) apoptosis were detected after NC, mimics and inhibitor transfection. (E) Cleaved caspase-3, Cleaved caspase-7, Cleaved caspase-9 and Cleaved PARP were detected by western blot. (F) Cell invasion and migration activity was assayed by transwell. (G) Cell morphology was photographed and confocal microscopy was used to test the E-cadherin and β-catenin expressions after transfections. (H) E-cadherin, Vimentin, N-cadherin and Fibronectin were analyzed by western blot. Data represent mean ± SD of three replicates. *P < 0.05.
