VE-PTP consists of multiple extracellular fibronectin III-like repeats (blue), a transmembrane domain and an intracellular protein tyrosine phosphatase domain (PTP). VE-cadherin consists of 5 extracellular cadherin domains (green), a transmembrane sequence and an intracellular cadherin cytoplasmic region (grey). The constructs VE-PTP-VN and VE-cadherin-VC have a modified N- or C-terminal sequence of the Venus protein (a yellow fluorescent protein (YFP) variant; represented by a half cylinder) inserted five residues after the transmembrane domain. A myc-tag (pink) has been inserted after the signal peptide of VE-PTP to facilitate analysis of expression. When brought into close proximity by a pair of interacting proteins, the fragments assemble to yield a fluorescent protein (BiFC signal).
