Compound 13d inhibits Tm-induced XBP1s mRNA expression in INS-1 cells. INS-1 cells were treated with or without Tm (0.1 µg/mL) in the presence of 13d or DMSO for 8 h. (A) XBP1s mRNA levels were analyzed by qRT-PCR. Relative mRNA levels were normalized against the housekeeping gene Cyclophilin A using the comparative CT method. ** P < 0.01 compared with Tm treatment alone. Bars indicate SD. (B) XBP1 mRNA levels were analyzed by RT-PCR and the products were resolved by agarose gel electrophoresis. The full-length (unspliced, XBP1u) and spliced (XBP1s) forms of XBP1 mRNA are indicated. Cyclophilin A mRNA was used as a loading control. Quantification was analyzed by ImageJ program shown at the bottom. The data shown are representative of 3 independent experiments.
