The activity of WNT/β-catenin pathway detected by western blot analysis and TOP/FOP flash experiments. The western blot results showed that overexpression of MEG3 resulted in decreased expression of β-catenin protein compared with the control group, so as to decrease the levels of downstream TCF-4 and cyclin D1 protein, while opposite results were found in cells with MEG3 silencing. The results of TOP/FOP flash experiments showed that the activity of luciferase was decreased by the transfection of MEG3 overexpression vector compared with the control group, while the activity of luciferase was increased after transfecting with MEG3 interference vector (*p<0.05).
