(A) CNE-1 and HNE-1 cells were seeded and transfected with negative control siRNA or DEPDC1 siRNA. Forty-eight hours after transfection, DNA biosythensis was labelled by BrdU (green). DAPI was used to stain cell nuclei (blue). BrdU positive cells were counted and subjected to statistical analysis (P* < 0.05, Student's t-test). Scale bar 50μm. (B) CNE-1 and HNE-1 cells were transiently transfected with siRNA as in Figure 4A. Forty-eight hours after transfection, cells were fixed and stained with anti-phospho-histone H3 antibody (green). The pHH3 positive cells were counted and subjected to statistical analysis (P** < 0.01, Student's t-test).
