Real time RT-PCR analysis and western blot analysis of the expression of indicated genes and proteins. (A, B) Human HaCat keratinocytes were cultured under normoxic (21% O2) and hypoxic (1% O2) conditions for 24 h, and harvested for RT-PCR and western blot analysis of RORα (A) or indicated genes (B). (C, D) Mouse PAM21 keratinocytes were cultured under normoxic (21% O2) or hypoxic (1% O2) conditions for 24 h, and harvested for RT-PCR and western blot analysis of RORα (C) or the indicated genes (D). The mRNA level of each gene was normalized to 36B4. The protein level of indicated genes was quantified by densitometry scanning, and normalized to α-tubulin.
