Ex vivo expanded primary human CD8+ CD62L+ T cells were lentivirally transduced with R12 or XBR1-402-derived CARs containing CD3ζ and 4-1BB signaling domains and a short IgG4 hinge-derived spacer. (A) CFSE-labeled CAR-Ts and untransduced T cells were incubated with the indicated target cell lines at an E:T ratio of 4:1 for 72 h and analyzed by flow cytometry. (B) CAR-Ts and untransduced T cells were incubated with the indicated target cell lines at an E:T ratio of 4:1 for 24 h. IFN-γ and IL-2 concentrations in the supernatant were measured by ELISA. (C) CAR-Ts and untransduced T cells were incubated with the indicated firefly luciferase-transduced target cell lines at the indicated E:T ratios for 7 h. Specific lysis was measured with a luciferase-based cytotoxicity assay.
