Schematic overview of the ubiquitylation step in US2-mediated HLA-I downregulation. US2 engages β2m-associated HLA-I and directs it to the E3 ubiquitin ligase TRC8. In cooperation with UBE2G2, TRC8 facilitates polyubiquitylation of HLA-I. Next, p97 catalyzes the extraction from the ER membrane into the cytosol for proteasomal degradation. UBE2J2 depletion increases TRC8 expression levels in the presence of US2, and in this way, enhances US2-mediated HLA-I downregulation.
