Role of NAPDH oxidase in Sap-induced netosis. Neutrophils (2.2 × 105) were incubated with the indicated Saps at a concentration of 1 ng/ml for 3 h to induce netosis. Cells with active NADPH oxidase (−DPI) or with this enzyme inhibited by 5 μM DPI (+DPI) treatment were used. The amount of released DNA was determined after SytoxGreen staining. Unstimulated neutrophils served as a negative control and 25 nM PMA treatment was assumed to induce a maximal level of NET release. Data are the means ± SEM from three experiments performed in duplicate. Asterisks denote statistical significance relative to the negative control (*p < 0.05, ***p < 0.005, ****p < 0.001, ns—not significant).
