Effects of Dkk1 silencing on TGF‐β1‐treated HSC‐T6 activation. After transfection with Dkk1 siRNA or scrambled RNAi, HSC‐T6 cells were exposed to TGF‐β1 for 2 days. (A) HSC‐T6 cells treated by TGF‐β1 with or without siRNA then total RNAs were isolated and real‐time PCR was performed to examine the mRNA levels of Dkk1 and α‐SMA. (B) HSC‐T6 cells treated by TGF‐β1 with or without siRNA then whole‐cell extracts were isolated and Western blotting was performed to examine the protein levels of Dkk1, β‐catenin, C‐myc and CyclinD1. (C) HSC‐T6 cells treated by TGF‐β1 with or without siRNA then whole‐cell extracts were isolated and Western blotting was performed to examine the protein levels of α‐SMA.
