15-LOX-2 reaction in intact cells. Ca2+ concentrations in the cell increase in response to stress. The Ca2+ binds to the PLAT domain of 15-LOX-2, resulting in a translocation from the cytosol to the membrane. 15-LOX-2 then binds its substrate, AA, and oxygenates it to form 15-HpETE. 15-HpETE is further metabolized by downstream enzymes to form lipoxins, which initiate the resolution of inflammation.
