Functional assay of the 5-FU patch in vitro. (a) The 265 nm UV absorbance of serially diluted 5-FU showed a linear correlation (R² = 0.9988). (b) The correlation between UV absorbance and drug concentration was assessed by collecting the media supernatants of the 5-FU patch at different time points. Most of the drug was released within 10 days, and drug release was continuous for up to 30 days. (c) The viability of BxPC3-luc cells was investigated using the media supernatants of the drug patches at the different time points. The cytotoxic effect of the released drug was observed predominantly in the early period, and the therapeutic effect was maintained for up to 28 days (*p < 0.05).
