(A) Outline experimental design for AML xenografts derived from OCI-AML3 cells transduced with firefly luciferase, selected, and drug treated for 24 hours with DMSO vehicle control, 1 μM Vorinostat, 2.5 μM of SANT-1 or combination treatment. Treated cells were transplanted into NSG mice (n=2) via tail vein. (B) Mice were imaged at days 15, 20 and 28 to assess cancer burden. Mice were sacrificed at the first sign of disease and survival extension was assessed. Average survival post-transplant for each treatment arm were as follows: D29.5 (DMSO), D30 (Vorinostat), D31 (SANT-1) and D36 (Vorinostat and SANT-1).
