(A) FLAG-ARD1 was immunoprecipitated from HEK293T cells, and co-precipitation of endogenous SAMHD1 was analyzed by western blot. (B) Recombinant GST-SAMHD1 proteins were subjected to in vitro acetylation assay with ±His-ARD1. The acetylation level was determined with anti-Lys-Ac antibody. (C) Lys-acetylated proteins were precipitated from GFP-mock or ARD1 expressing HEK293T cells and were blotted with anti-SAMHD1 antibody. (D) FLAG-ARD1 wildtype, K136R and DN mutants were overexpressed in HEK293T. Lysed proteins were precipitated with anti-Lys-Ac antibody and were blotted with anti-SAMHD1 antibody.
