ROS-mediated inhibition of cytokine production imparts a selective role for STIM2 in neutrophils. (A) Analysis of TNFα production in WT neutrophils treated with the NADPH oxidase inhibitor DPI. Bone marrow cells (1 × 106) from WT mice were incubated with zymosan (100 μg/mL) for 5 hours with or without DPI (10 μM), and TNFα levels were measured by intracellular staining. (B) Analysis of NF-κB activation in WT neutrophils treated with DPI. Isolated neutrophils were stimulated with zymosan (100 μg/mL) in the presence or absence of DPI (10 μM), and nuclear and cytoplasmic fractions were blotted for the indicated proteins.
