An EP300 chemical inhibitor suppresses growth of lung cancer cell lines with 3q gains and increases sensitivity to a PI3K inhibitor. A, Single agent EP300 chemical inhibitor treatment suppresses growth of lung cancer cell lines with 3q gains. Cells were fed every other day with the indicated CBP30 concentration and growth quantified after 7–10 days by alamarBlue. Data are plotted relative to control vehicle-treated cells, which was assigned a value of 100. Means ± S.E. from triplicate cultures are shown. Significance was calculated using a 2-tailed t test. Unless otherwise indicated, p values are relative to respective DMSO-treated control cultures. MGH7, ***p = 0.0001 (10 μm CBP30), 1.9 × 10−6 (20 μm CBP30); ChaGoK1, ***p = 3.5 × 10−6 (10 μm CBP30), 1.4 × 10−5 (20 μm CBP30); SW1573, **p = 0.0007 (10 μm CBP30), 0.001 (20 μm CBP30); ***p = 7.1 × 10−6 (20 μm CBP30/MGH7 versus 20 μm CBP30/SW1573), 6.7 × 10−5 (20 μm CBP30/ChaGoK1 versus 20 μm CBP30/SW1573). B, At low doses, combination of PI3K and EP300 chemical inhibitors results in more growth inhibition than either single agent alone. Growth assays were performed as described in (A). Significance was calculated using a 2-tailed t test. ns = not significant. MGH7, **p = 0.001, ***p = 5 × 10−5; ChaGoK1, *p = 0.009, **p = 0.005, SW1573, ns, p = 0.40 (to CBP30 alone), 0.17 (to BKM120 alone).
